Effects of prepregnancy obesity and excessive gestational weight gain on iron-related indicators of offspring rats

摘要目的 探究母鼠孕前肥胖和孕期过度增重对子鼠离乳后铁相关指标的影响及可能机制。方法 将18只健康的SPF级Wistar雌性大鼠以1∶1∶1比例随机分为对照组(n=6)、孕前肥胖组(n=6)和孕期过度增重组(n=6),对照组全程饲喂标准饲料,孕前肥胖组于孕前6周起饲喂高脂饲料,孕期过度增重组仅于孕前10 d起饲喂高脂饲料、其余时期饲喂标准饲料。所产子鼠于哺乳期第21天离乳,36只子鼠(每组12只)纳入离乳后研究,雌雄各半,饲喂标准饲料;子鼠离乳后第7天、14天和21天检测血清/全血炎性指标和铁相关指标,前者包括白细胞介素-6(IL-6)和铁调素,后者包括铁蛋白、血红蛋白(Hgb)、平均血红蛋白量(Mch)和平均红细胞体积(Mcv)。结果 孕前肥胖组母鼠孕前体重显著高于对照组[(327.7±27.0 )g vs. (279.3±14.2)g,P<0.01],孕期过度增重组母鼠孕期增重均值超过对照组24.4%[(108.2±17.9)g vs. (87.0±12.0)g]。与对照组子鼠相比,孕前肥胖组和孕期过度增重组子鼠离乳后第7、14和21天铁蛋白水平显著升高,孕前肥胖组雌性子鼠离乳后第7天Mch水平偏低[(21.2±0.7) pg vs. (22.5±0.7)pg,P<0.05],孕期过度增重组雌性子鼠离乳后第7天Hgb[(96.3±3.9)g/L vs. (103.3±2.7)g/L]和Mch水平[(21.2±1.1) pg vs. (22.5±0.7)pg]偏低(P<0.05);未见孕前肥胖与孕期过度增重对雄性子鼠铁相关指标的显著影响。调整炎性指标后,母鼠孕前肥胖与孕期过度增重对子鼠铁蛋白和Hgb的影响不再具有统计学意义。结论 母鼠孕前肥胖和孕期过度增重影响雌性子鼠离乳后铁相关指标,炎症反应是可能机制之一。
Abstract:Objective To investigate the effects and associated mechanisms of prepregnancy obesity and excessive gestational weight gain on the iron-related indicators of offspring rats after weaning. Methods A total of 18 healthy female SPF Wistar rats were randomly divided into control group (Con, n=6), pre-pregnancy obesity group (Pre-obese, n=6), and excessive gestational weight gain group (GWG, n=6) in a 1∶1∶1 ratio. The Con group was treated with a standard chow diet throughout the study, while the Pre-obese group with a high-fat diet starting from 6 weeks before gestation, and the GWG group with a high-fat diet only from 10 days before gestation and a standard chow diet for the rest of the period. Their offspring rats were weaned on the 21st day of lactation. A total of 36 offspring rats (12 in each group) were included in the final analysis. The ratio of male and female rats was 1∶1 and all offspring rats were treated with a standard chow diet. The serum/whole blood inflammatory [interleukin-6 (IL-6) and hepcidin] and iron-related indicators [ferritin, hemoglobin (Hgb), mean corpuscular hemoglobin (Mch), and mean corpuscular volume (Mcv)] were measured on the day 7, 14, and 21 after weaning. Results The prepregnancy weight of the female rats in the Pre-obese group was significantly higher than the Con group [(327.7±27.0) g vs. (279.3±14.2) g, P<0.01]. The mean of gestational weight gain of the GWG group was 24.4% over the Con group [(108.2±17.9) g vs. (87.0±12.0) g]. Compared with the Con group, the level of ferritin of offspring rats in the Pre-obese group and GWG group was significantly higher on the day 7, 14, and 21 after weaning. The Mch level in the female offspring rats of the Pre-obese group was lower than the Con group 7 days after weaning [(21.2±0.7) pg vs. (22.5±0.7) pg,P<0.05], while both the Hgb and Mch levels of the GWG group were lower [(96.3±3.9) g/L vs. (103.3±2.7) g/L and [(21.2±1.1) pg vs. (22.5±0.7) pg, P<0.05]. However, the difference in iron-related indicators of the male offspring rats was not significant. After adjusting the levels of inflammatory indicators, it is worth noting that the effects of prepregnancy obesity and excessive gestational weight gain on the levels of ferritin and Hgb were not statistically significant anymore. Conclusion The prepregnancy obesity and excessive gestational weight gain adversely affected the level of iron-related indicators in female offspring rats after weaning. In addition, inflammation might play a partial role in the mechanism.
