Abstract:Objective To explore the role of Wnt7a in the epileptogenesis by the regulation of neurogenesis in hippocampus through observation Wnt7a mRNA and protein during the process of epileptogenesis in the Kainate-induced temporal lobe epilepsy model.Methods Animal model of temporal lobe epilepsy were Male Sprague-Dawley rats (Bought for the Second Affiliated Hospital of Harbin Medical University) which induced by kainic acid (KA) at postnatal day 30, Real-time PCR method and Western blot were performed to detect Wnt7a mRNA and protein in the hippocampus of experimental and Normal Saline control groups (NS group) at different stages of epileptogensis, the acute stage, the initial stage of spontaneous recurrent seizures (SRS) and the stable stage of chronic epilepsy.Results Wnt7a mRNA decreased at the acute stage (0.304±0.113,P<0.001), increased at the initial stage of SRS(3.369±1.455,P=0.010)and at the stable stage of chronic epilepsy (5.845±3.524,P=0.020) compared with the NS group (1.000±0.000), with statistical significance. For the KA group at different stages, the expression of Wnt7a mRNA elevated significantly both at the initial stage and chronic stage (P<0.05) in comparison with that at the acute stage. At the chronic stage, the expression of Wnt7a mRNA increased but the difference was not statistically significant (P>0.05) when compared with that at the initial stage. There was no significant difference of Wnt7a protein expression in the hippocampus in the time courses and the different groups.Conclusion Wnt7a may be involved in the epileptogenesis by the regulation of neurogenesis in hippocampus in kainate-induced epilepsy model.
