Abstract:Objective To discuss the effect of retinoic acid(RA) on proliferation and differentiation of cryptorchidism spermatogenic cells via miR-210.Methods To establish a model of flutamide-induced cryptorchidism mouse. The experiment was divided into normal control group, flutamide group, RA treatment group, miR-210 knockdown group, miR-210 overexpression group and miR-210 overexpression + RA treatment group. RT-qPCR was performed to assess the expression level of miR-210, C-kit and PLZF mRNA. Western blot was employed to detect the expression level of C-kit and PLZF protein.Results RA significantly upregulated the expression levels of C-kit and PLZF mRNA and protein in cryptorchidism tissues, and downregulated the expression levels of miR-210. Knockdown of miR-210 could upregulate the expression of C-kit and PLZF mRNA and protein in cryptorchidism tissues, while overexpression of miR-210 had the opposite result. At the same time, miR-210 overexpression + RA treatment could restore the inhibition of miR-210 overexpression on the expression of C-kit and PLZF.Conclusion miR-210 is highly expressed in cryptorchidism tissues, and RA can promote the proliferation and differentiation of cryptorchidism spermatogenic cells by downregulating the expression level of miR-210.
