Abstract:Objective To assess the effects of stachydrine, choline, and their combination from Leonurus japonicus Injection on uterus of rats.Methods Thirty SD rats were randomly divided into two groups:normal rat isolated uterus model (n=15) and spasmodic uterine model (n=15). The model uteruses were successively given stachydrine (40 μl, 40 μl, 80 μl), choline (40 μl, 40 μl, 80 μl), or their combination [40 μl(stachydrine of 20 μl+ choline of 20 μl), 40 μl (stachydrine of 20 μl+ choline of 20 μl), 80 μl (stachydrine of 40 μl+ choline of 40 μl)] that isolated from Yimucao Injection total alkaloids by way of cumulative dosing. Routine physiological recording and recording for the uterine contraction curve were performed, and the change of uterine contraction vitality, amplitude, and frequency were recorded before and after administration.Results Compared with pre-administration, administration of stachydrine, choline alone, and their combination had no significant effect on contractility activity, contractility tension, minimum tension value, and contraction frequency in the normal uterus model. In addition, the inhibition rate decreased with the increase of dose. However, the uterine contraction activity, contraction tension and frequency were significantly increased in the isolated uterus model, and the spasm was observed in vitro after stimulation with stachydrine and choline alone. The contraction tension and frequency of uterine smooth muscle were significantly lower than those of the model group, and the minimum value of systolic tension was significantly higher than that of the model group.Conclusion Stachydrine and choline on uterine activity showed two-way adjustment effects depending on the state of the uterus. Stachydrine and choline had no significant synergistic or antagonistic effect, but both were effective substances in Leonurus japonicus Injection.
