The levels of 8-OHdG and 4-HNE in follicular fluid among patients with decreased ovarian reserve function and their relationship with in vitro fertilization-related indexes

Abstract:Objective To investigate the levels of 8-hydroxydeoxyguanosine (8-OHdG) and 4-hydroxynonenal (4-HNE) in follicular fluid among patients with decreased ovarian reserve function (DOR) and their relationship with related indexes of in vitro fertilization (IVF).Methods The follicular fluid of patients undergoing IVF-assisted pregnancy in the Department of Reproduction Medicine of Shanghai First Maternity and Infant Hospital from July 2020 to July 2021 were collected for analysis. Among them, 37 patients with DOR infertility were included in the DOR group, and 42 women with normal ovarian reserve function (NOR) due to fallopian tube factors were selected as control group. The levels of 8-OHdG, 4-HNE and the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) were compared between the two groups, and the relationship between 8-OHdG, 4-HNE and IVF-related indexes was analyzed.Results Compared with the control group, the number of oocytes obtained, the number of MⅡ, the number of fertilization and high-quality embryos in DOR group were significantly decreased (P<0.05). The levels of 8-OHdG and 4-HNE in follicular fluid of patients with DOR group were significantly increased, while the activities of SOD, GSH-Px and CAT were significantly decreased (P<0.05). Correlation analysis results showed that the levels of 8-OHdG and 4-HNE in follicular fluid of patients with DOR group were positively correlated with age and basal FSH (r>0, P<0.05), while 8-OHdG was negatively correlated with AMH, number of fertilization and high-quality embryos (r<0, P<0.05). 4-HNE was negatively correlated with AMH, MⅡ number and number of fertilization (r<0, P<0.05).Conclusion Oxidative stress reaction exists in follicular fluid of DOR patients, which may affect the maturation, fertilization and embryonic potiential development, and then affect the outcome of IVF pregnancy in DOR patients.
