The relationship between the expression of LncRNA HIF1A-AS1 and HIF1A in placental tissues with the recasting of placental spiral artery in late onset severe preeclampsia

Abstract:Objective To analyze the relationship between the abnormal expression of long non-coding RNA hypoxia-inducible factor 1 alpha-antisense RNA 1 (LncRNA HIF1A-AS1) and hypoxia-inducible factor-1 alpha (HIF1A) in placental tissues with the placental spiral artery recasting of pregnant women with late onset severe preeclampsia. Methods We selected ninety-two pregnant women with late onset severe preeclampsia who gave birth in our hospital from October 2019 to April 2021 as the preeclampsia group, and 86 pregnant women with normal pregnancy during the same period as the normal pregnancy group. We also collected the general data about age, systolic blood pressure and diastolic blood pressure of each pregnant woman. Real-time fluorescent quantitative PCR (qRT-PCR) method was used to detect the levels of LncRNA HIF1A-AS1 and HIF1A mRNA in the placental tissue, and scanning electron microscope was used to measure the lumen area and wall thickness of the placental spiral artery. We used Pearson′s method to analyze the correlation between LncRNA HIF1A-AS1 and HIF1A mRNA levels with different parameters in the placental tissues of pregnant women with late onset severe preeclampsia. Results Compared with the normal pregnancy group, the level of HIF1A mRNA in the placental tissue, average wall thickness, systolic blood pressure and diastolic blood pressure were significantly increased in the preeclampsia group. The LncRNA HIF1A-AS1 level and the average lumen area were significantly decreased (P<0.05). There was a negative correlation between LncRNA HIF1A-AS1 and HIF1A mRNA levels in the placental tissue of pregnant women with late onset severe preeclampsia (r=-0.628, P<0.05). LncRNA HIF1A-AS1 was positively correlated with the average lumen area (P<0.05), and was negatively correlated with the average tube wall thickness, systolic blood pressure and diastolic blood pressure (P<0.05). HIF1A mRNA level was negatively correlated with the average lumen area (P<0.05), and positively correlated with the average tube wall thickness, systolic blood pressure and diastolic blood pressure (P<0.05). Conclusion The expression of LncRNA HIF1A-AS1 and HIF1A is abnormal in the placental tissue of pregnant women with late onset severe preeclampsia. It may be related to insufficient recasting of the placental spiral artery.
