Abstract:Objective To investigate the expression changes of matrix metalloproteinases (MMPs) in the villus tissue of patients with missed abortion, and to explore its role and regulatory mechanism in the missed abortion.Methods 30 patients with missed abortion were selected as the research group, and 30 healthy and abortion women in early pregnancy were selected as the control group from November 2015 to May 2016 in the family planning clinic of the First Affiliated Hospital of Soochow University and the Department of Obstetrics and Gynecology, North District of Suzhou Municipal Hospital. The level of TGF-β1 in plasma was measured by ELISA. MRNA expressions of NF-κB, TGF-β1, TIMP-1 and MMP-9 in villus tissue were detected by RT-PCR,and the protein levels of NF-κB, TGF-β1, TIMP-1 and MMP-9 were detected by Western blotting.Results (1)The average level of TGF-β1 in plasma of the research group was lower than that of the control group (P<0.05).(2)The mRNA expression levels of NF-κB1, TGF-β1 and TIMP-1 in villus tissue of the research group were lower than those of the control group, and the expression level of MMP-9 mRNA was higher than that of the control group (P<0.05).(3)The expression level of MMP-9 protein in villus tissue of the research group was higher than that in the control group, and the levels of NF-κB p50, TGF-β1 and TIMP-1 protein were lower than those of the control group(P<0.05).Conclusion The decreased expression level of NF-κB can affect the expression level of TGF-β1, and it may influence the balance of MMP-9/TIMP-1 and lead to the excessive degradation of extracellular matrix, and result in the failure of embryo implantation and impaired placenta formation, eventually cause the termination of embryo development.
