Non-targeted metabolomics study of the discordant growth of dichorionic twins

Abstract:Objective To explore the difference in the metabolic profile of the cord blood of the two fetuses with discordant growth in dichorionic twins with non-targeted metabonomics technology.Methods Twenty-six pregnant women who met the diagnostic criteria for discordant growth of dichorionic twins were enrolled. Ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) was used to detect metabolites in the cord blood of twin fetuses. Partial least squares discrimination analysis (PLS-DA) were used to perform multivariate pattern analysis on mass spectrometry data, and the variable weight value (VIP) was calculated according to the PLS-DA model. Combining with univariate statistical analysis Wilcoxon rank sum test and fold change (FC) analysis, we used VIP>1, FC>1.2 or FC<0.83 as the screening condition to screen the characteristics of different metabolites, and conducted pathway analysis.Results In twins with discordant growth, a fetus whose weight is less than the 10th percentile for the same gestational age suggests that the fetus has growth restriction. In the same maternal environment, the characteristics of cord blood metabolites of growth-restricted infant and normal fetuses were different. A total of 35 different metabolites were identified in the comparison between the two groups. Among them, 10 metabolites such as methionine, trans-2-dodecanoylcarnitine, 17α,21-dihydroxypregnenolone and prostaglandin D3 were up-regulated in growth-restricted infant. Choline, arginine and other metabolites decreased. Pathway analysis showed that the identified different metabolites have important effects on methionine metabolism, spermine metabolism, betaine metabolism and glycerophospholipid metabolism. Conclusion The cord blood of the two fetuses with discordant growth in dichorionic twins underwent metabonomic changes, which were mainly manifested as metabolic abnormalities in amino acids and lipids.
