Abstract:Objective To investigate the expression of cytoplasmic polyadenylation element binding protein 4 (CPEB4) in cervical cancer and its effect on proliferation, migration and invasion of cervical cancer Siha cells.Methods We collected cancerous tissues and normal adjacent tissues of 42 patients with cervical cancer. Quantitative real-time PCR and Western blot were performed to detect mRNA and protein levels of CPEB4 in cervical cancer and pericarcinomatous tissues. CKK-8 assay, transwell assay and western blot were used to evaluate cell proliferation, migration and invasion, and protein expression of P53 and Twist1 after transfection of CPEB4 siRNA in cervical cancer Siha cells.Results Compared with pericarcinomatous tissues, the mRNA and protein expression of CPEB4 were both significantly up-regulated in cervical cancer tissues. The proliferation activity of Siha cells transfected with CPEB4 siRNA was weakened, and the migration and invasion ability were significantly decreased (P<0.05). Moreover, the protein expression of P53 was significantly up-regulated and Twist1 was significantly down-regulated in Siha cells transfected with CPEB4 siRNA (P<0.05).Conclusion The high expression of CPEB4 in cervical cancer may affect the proliferation, migration and invasion of cervical cancer cells by regulating the expression of P53 and Twist1, and CPEB4 may be a diagnostic marker and potential therapeutic target for cervical cancer.
