The potential effect of BMP15 and BMPR II abnormal expression on granulosa cell proliferation in patients with PCOS

Abstract:Objective To investigate the potential effect of BMP15 and BMPR II abnormal changes on IVF outcome through the granulosa cell apoptosis.Methods A total of 138 PCOS patients (PCOS group) and 78 female patients with normal ovarian function (control group) were included. General information, retrieved oocyte number, BMI, Gn dosage, menarche age, fertilization rate, portable embryo rate, high-quality embryo rate, and clinical pregnancy rate were recorded. Real-time PCR was used to detect the mRNA levels of BMPR II, Bcl-2, and Caspase-3 in granulosa cells. Western blotting was used to detect the expression of BMP15 in the follicular fluid. Apoptosis index of granulosa cell was detected by flow cytometry.Results Compared with the control group, age, BMI, portable embryo rate, and serum FSH levels were significantly decreased in the PCOS group. BMP15 expression was significantly decreased in PCOS group when compared with the control group, whereas, the expression of BMPR II was significantly increased. The apoptosis index was significantly increased in the PCOS group compared with the control group.Conclusion Abnormal changes of BMP15 and BMPR II may affect oocyte quality and embryo development potential, and these changes were correlated with granulosa cell apoptosis.
